Heterocyclic compounds serve as crucial structural elements in the field of pharmaceutical medicinal chemistry. The five-membered heterocyclic nucleus of 1,3,4-oxadiazole occupies a distinctive position within medicinal chemistry and plays a vital role in the development of anticancer agents. Recognized as a significant pharmacophore for approximately 85 years, 1,3,4-oxadiazole is in high demand across various biological and chemical disciplines. This small and straightforward nucleus is found in numerous compounds that are the focus of research concerning their properties, synthesis, derivatives, and pharmacological activities, including anticancer, antibacterial, antimalarial, anti-inflammatory, antidepressant, analgesic, and antiviral effects. The information regarding these activities presented in this article may prove beneficial to researchers, facilitating the discovery of new therapeutic agents for the benefit of society.
Objective: This study involved the synthesis of new Schiff bases and 1,3-oxazepine derivatives from the baclofen drug and study the anticancer activities. Methods: Baclofen was initially reacted with aromatic aldehydes to create Schiff base derivatives (Ia–Ib), which were then closed in the next step using anhydrous acids to form oxazepine derivatives (IIa–IId). Results: The title compounds were synthesized successfully and identified using FT-IR, 1H NMR, and 13C NMR spectroscopy. Characterization of these compounds has been done on the basis of FT-IR and UV-Vis, as well as magnetic susceptibility and conductivity measurements. Abstract As a part of our ongoing project on the design and synthesis of new 4-thiazolidinone derivatives with antimicrobial activity, four new 4-thiazolidinone derivatives carrying bromo, nitro, methyl, and chloro groups on the benzene ring were synthesized by starting with the 7-amino-4-methylcoumarin moiety, linking coumarin with various phenyl isothiocynate to form the thiourea group, and then cyclizing the derivatives, characterized by IR and 1HNMR, and assayed in vitro for their antimicrobial activity against Gram positive and Gram negative bacteria and fungi.
