Heterocyclic compounds serve as crucial structural elements in the field of pharmaceutical medicinal chemistry. Recognized as a significant pharmacophore for approximately 85 years, 1,3,4-oxadiazole is in high demand across various biological and chemical disciplines. This small and straightforward nucleus is found in numerous compounds that are the focus of research concerning their properties, synthesis, derivatives, and pharmacological activities, including anticancer, antibacterial, antimalarial, anti-inflammatory, antidepressant, analgesic, and antiviral effects. The information regarding these activities presented in this article may prove beneficial to researchers, facilitating the discovery of new therapeutic agents for the benefit of society.
In the present study, chalcone derivatives were synthesized via the Claisen-Schmidt condensation of 2-methyl acetanilide or 4-nitro acetanilide with p-dimethyl amino benzaldehyde in an ethanolic sodium hydroxide solution. The resulting chalcone reacted with urea or thiourea to produce several novel pyrimidine derivatives. All the synthesized compounds were characterized by FTIR, 1HNMR and 13CNMR spectroscopy. Before the synthesis process, molecular docking studies were performed to evaluate the potential of the synthesized molecules as drug candidates. Reviewing these studies, we found that uracil can be coordinated through the electron pair on the N1, N3, O2, or O4 atoms. If the uracil was a mono-dentate ligand, it will be coordinated by one of the following atoms: N1, N3 or O2. But if the uracil was bi-dentate ligand, it will be coordinated by atoms N1 and O2, N3 and O2 or N3 and O4. However, when uracil forms complexes in the form of polymers, coordination occurs through the following atoms: N1 and N3 or N1 and O4.

The nature of bonding and the stereochemistry of the complexes were deduced from metal analyses, infrared, electronic spectra,magnetic susceptibility and conductivity measurements, an octahedral geometry was suggested for all complexes except the copper complex has a square planar geometry .preliminary in vitro tests for antimicrobial activity show that all the prepared compounds except iron complex display good activity to gram positive Staphelococcus aures and gram negative Escherchia coli.
