Application of RAPD-PCR and Phylogenetic Analysis for Accurate Characterization of Salmonella spp. Isolated from Chicken and Their Feed and Drinking Water

The aim of this study was ‎the‎ discrimination of Salmonella‎‎ isolated from chicken and their feed ‎and drinking water for the epidemiological control of salmonellosis. Totally, 289 samples, ‎including 217 chicken cloaca swabs, 46 water, and 26 feed samples were collected from five ‎different farms in Karbala governorate, Iraq. Conventional bacteriology tests, API 20E, Vitek 2, ‎and serology were used for bacterial identification. Random amplified polymorphic ‎DNA (RAPD)-polymerase chain reaction (PCR) was applied to analyze the genetic relationships ‎among Salmonella‎‎ isolates. The isolation rate of Salmonella‎‎ spp. was 21.1% (61/289). While the ‎water samples constituted the highest rate (30.4%), a rate of 21.7% was reported for the cloaca ‎swabs, with no isolate at all from chicken feed. Vitek 2 was able to identify some isolates to the ‎serotype level, such as S. Enteritidis, S. Paratyphi B, and S. Paratyphi C. However, the isolates ‎were diagnosed as S. enterica by API 20E, and as S. enterica subsp. arizonae through serology. ‎Analyzing the samples by the RAPD-PCR assay showed the presence of genetically different ‎Salmonella‎‎ spp. Dendrograms created by the GelJ software successfully delineated the genetic ‎relationships. Therefore, RAPD-PCR can be used as a surrogate tool for the fast, reliable, and ‎accurate detection of Salmonella‎‎ in epidemiological surveys when compared with other ‎biochemical-based identification methods.

The current study aimed to isolate and diagnose Candida spp yeasts that cause candidiasis with a PCR device from patients reviewed for some hospitals in Baghdad city and by 190 samples, the study recorded 123 isolates and the total percentage of infection was 64.7% .Samples were taken from different clinical cases of the vagina, blood and mouth and the Candida spp were (70.37%, 41.26%, 86.95%) respectively. The Fluoride Concentration of Bottled Drinking Water in Al-Basra City, Iraq

Background: This study aimed to determine the amount of fluoride in commercially available bottled drinking water in Al-Basra city, Iraq Materials and Methods: Eleven brands of bottled drinking water were obtained from supermarkets in Al-Basra city, Iraq. Five samples of 10 ml. were taking from each one of brands and the fluoride was determined by using fluoride ion selective electrode. Results: The highest fluoride concentration was present in BADIOT brand (1.174 mg/L) while the lowest was in Barakat brand (0.038 mg/L). Isolation of Candida Spp. from Patients with Different Types of Leukemia Who Suffered Oral Candidiasis Due to Their Weekend Immune System

The study is concern on determine the type of Candida spp.in leukemia patients that were infected with oral candidiasis as a result to their immune suppression (weekend immune system) due to their submission to radiation and chemotherapy treatment. The result showed that the most common isolates were C. guillermondii 19 which represent 31.66% of cases, then followed by C. itermedia 11 which represent 18.3%, while the less common isolates were for C. zeylamodes 3 which represent 5%.
