Targeted Intranasal Delivery of Antiretrovirals for NeuroAIDS: Opportunities, Barriers, and Formulation Strategies

NeuroAIDS refers to the spectrum of neuropsychiatric complications observed in approximately 30–50% of HIV/AIDS patients. Although the advent of antiretroviral therapy has reduced the severity of AIDS, mild cognitive deficits persist in nearly half of treated patients, highlighting limitations in central nervous system (CNS) drug delivery. These limitations are attributed to poor blood-brain barrier (BBB) permeability, P-glycoprotein efflux, low solubility, first-pass metabolism, and short half-lives of many ART drugs. Nose-to-brain drug delivery has emerged as a promising alternative, bypassing the BBB via olfactory and trigeminal pathways. This route offers direct CNS access while minimizing systemic exposure. Factors such as drug molecular weight, lipophilicity, pH, osmolality, viscosity, particle size, and surface charge significantly influence nasal drug absorption and brain targeting. Approaches to optimize nose-to-brain delivery include the use of prodrugs, permeation enhancers, mucoadhesive agents, enzyme inhibitors, and P-glycoprotein inhibitors. Despite encouraging preclinical data, challenges remain, including mucociliary clearance, enzymatic degradation, and species-specific anatomical variations that hinder translational research. A rational formulation design that considers physiological and physicochemical barriers is essential for clinical success. Nanocarriers—including nanovesicles, solid lipid nanoparticles (SLNs), and microemulsions—offer several advantages: they protect drugs from enzymatic degradation, improve solubility and stability, enable controlled release, and enhance mucoadhesion and uptake across the nasal mucosa. Overall, intranasal delivery systems, particularly those employing nanocarriers and functional excipients, offer a novel and efficient approach to managing NeuroAIDS by enhancing ART bioavailability in the CNS and improving drug access to HIV reservoirs in the brain.

Formulation and In Vitro Evaluation of Mucoadhesive Antimicrobial Vaginal Tablets of Ciprofloxacin Hydrochloride

Ciprofloxacin is a broad spectrum fluoroquinolone, effective in the treatment of a wide range of infections, including genitourinary tract infections.In this study, bioadhesive vaginal tablets of ciprofloxacin hydrochloride were prepared by direct compression method using a combination of bioadhesivepolymers carbopol 934P(Cp), carboxymethylcellulose (CMC) and sodium alginate (SA) in different ratios.The prepared tablet formulations were characterized by measuring their swelling capacity, surface pH, bioadhesive properties, and in-vitro drug dissolution. It was found that the bioadhesive force was directly proportional to carbopol 934P content in different formulae and was further enhanced by the inclusion of carboxymethylcellulose.

In Vitro Study of MefenamateStarch as Drug Delivery System

Mefenamic acid was esterified with starchwith[1:1] Molar ratio, as drug substituted with natural polymer, to prolongthe period of hydrolysis of drug polymer with other advantages. The new prodrug starch was characterized by FT-IR and UV-Visible and 1H-NMR spectroscopies. The physical properties were studied and controlled drug release was studied in different pH values at 37oC. The stability of drug was carried out by measuring the absorbance of mefenamic starch which hydrolyzed in HCl solution of pH 1.1 (artificial gastric fluid) and phosphate buffer of pH 7.4 (simulating intestinal fluid SIF) at 37oC for several days. The Morals Strategies from Sunnah Depending

The study aims at elicitation the morals strategies from sunnah depending on the analytic-qualitative method. It revealed a comprehensive integrated system of creative strategies to improve morals. It was categorized into three areas: cognitive strategies, affective strategies, and some strategies based on self-initiative and practice. The researcher recommended that teachers and educators should employ these effective strategies in the process of moral guidance for the Muslim generation. He also recommended that researchers should put more effort to do more In-depth qualitative studies, which must be directed to establish the origin of different education strategies from the Holy Qur’an, the Sunnah, and Muslim scholars’ legacy.
