Optimization And Development Of Ondansetron HCl Nanocrystals By Using D-Optimal Design Expert® And In-Vitro Characterization

Background: Over the last few years, nanocrystal technology has expanded to improve the bioavailability of poorly water-soluble drugs, which is challenging. Nanocrystals (NCs) exhibit many properties, such as enhancing drug solubility, dissolution, oral absorption, and high drug loading. Ondansetron hydrochloride (ONH) is an antiemetic drug that antagonizes a serotonin 5-HT3 receptor in the peripheral and central nervous system and is widely prescribed for the management of chemotherapy and radiotherapy-induced vomiting and nausea, as well as for postoperative nausea. ONH exhibits approximately 60% bioavailability due to its poor solubility and first-pass metabolism in the liver. Its solubility is pH-dependent, its precipitates above pH 6. Objective: Preparation of ONH-NCs to increase its dissolution rate as a preliminary study to be prepared as a sublingual film to avoid first-pass metabolism. Method: NCs were prepared using the nanoprecipitation method and optimized via a D-optimal surface design considering drug to stabilizer ratio, solvent to anti-solvent ratio, stirring rate, and stabilizer type. Results: The process yielded particle sizes (PS) ranging from 99-409 nm and a polydispersity index (PDI) range of 0.08-0.44 using stabilizers such as Soluplus®, Brij 35®, and Kollidon VA 64® (Koli 64). Differential scanning calorimetry (DSC) and X-ray diffraction (XRD) demonstrated showed reduced crystallinity, while Fourier transform infrared (FTIR) spectroscopy confirmed drug-stabilizer compatibility, and Field Emission Scanning Electron Microscopy (FESEM) depicted its morphological characterization. Conclusion: The results revealed that nanocrystallization of ONH may enable faster drug release at pH 6.8.

Treatment of shallow and deep white spot lesions with three different mouthwashes evaluated by laser fluorescence (an in vitro study)

This research aims to find how three different types of mouthwashes affect the depth of artificial white spot lesions. Teeth with various depths of white spot lesions were immersed in either splat mouthwash, Biorepair mouthwash, Sensodyne mouthwash, or artificial saliva (control)twice daily for one minute for 4 weeks and 8 weeks at 37°C. After this immersion procedure, lesion depth was measured using a diagnosed pen score. A one-way analysis of variance, Dunnett T3 and Tukey's post hoc α = .05 were used to analyze the testing data. Orange peel was used as a plant-derived medium to prepare a ZnO–calcite mixed-phase nanostructured material, which was physicochemically characterized and evaluated for DPPH radical-scavenging, α-glucosidase inhibitory, and in vitro cytotoxic activities. XRD confirmed the coexistence of hexagonal wurtzite ZnO and crystalline calcite, with ZnO apparent crystallite sizes ranging from 9.2 to 28.3 nm (mean 16.4 ± 7.3 nm). Study of effect acidic solution (HCl) and (EP/Al2O3 & EP/ TiO2) hybrid on thermal conductivity of epoxy resin.

This research studies the effect of adding micro, nano and hybrid by ratio (1:1) of (Al2O3,TiO2) to epoxy resin on thermal conductivity before and after immersion in HCl acid for (14 day) with normality (0.3 N) at weight fraction (0.02, 0.04, 0.06, 0.08) and thickness (6mm). The results of thermal conductivity reveled that epoxy reinforced by (Al2O3) and mixture (TiO2+Al2O3) increases with increasing the weight fraction, but the thermal conductivity (k) a values for micro and Nano (TiO2) decrease with increasing the weight fraction of reinforced, while the immersion in acidic solution (HCl) that the (k) values after immersion more than the value in before immersion.
