Genomic and Metabolic Adaptations of HydrocarbonDegrading Bacteria in Iraq’s Surface Waters

Oil exploration and extraction operations have contributed significantly to hydrocarbon contamination in surface water in Iraq. Molecules which acquire an adaptive properties through the genomic and metabolic changes of indigenous bacteria able to live in hydrocarbon polluted areas of the world are investigated. Water samples were gathered from twelve distinct polluted locations. GC-MS was used to determine the amount of hydrocarbons in the water. Chemical oxygen demand (COD) was measured to determine the level of pollution. Citrobacter freundii and Bacillus cereus were isolated and identified using 16S rRNA sequencing and VITEK 2. Bioinformatics analysis and whole-genome sequencing (WGS) were used to identify genetic mutations in hydrocarbon-degrading pathways. Metabolomic and proteomic analyses assessed functional adaptations. Phylogenetic analysis showed that these strains are genetically related to industrial and coastal bacteria from China and India. Statistical analyses of variance (ANOVA) and Pearson correlation were performed to verify that the results were robust. This study provides valuable insights into microbial adaptation in hydrocarbon-impacted habitats, and assesses the suitability of such strains in bioremediation technologies. ABSTRACT The isolation and characterization of (27) isolate of extreme halophilic bacteria was performed ninteen isolate belonged to the genus Halobacterium which included Hb.halobium. Hb. salinarium, Hb. volcanii. Growth curve and generation time in logarthmic phase was measured and found to be (12.8hr±0.32), (11.2hr±0.2), (9.8hr±0.87), respectivaly. Effect of various concentrations of NaCl, KCI, NH4Cl and MgSO4.7H2O was studied, NaCl was essential for the rod shape rapid growth Rat and pigmentation. Less than 1% concentration caused lysis of bacteria. Yeast extract was the best carbone source as compared with glucose and casamino acid.

The effect of methanol and water plant extracts of Cordia myxa on Some pathogenitic bacteria and Candida albicans

this study aimed to study the effect of Cordia myxa extract on the growth and activities of the following types of bacteria : Escherichia coli , Pseudomonas aeruginosa, Proteus Spp., Klebsiella pneumoniae , Staphylococcus aureus, Streptococcus pyogenes , Bacillus subtilus, and the yeast Candida albicans .the results showed an inhibitory effect of the methanol extract on both the growth and activity of the tested microbes .this was reflected by the minimum inhibitory concentration ( MIC ) of different type of bacteria and the yeast.
