Background: The beneficial gut bacterium E. coli can cause blood poisoning, diarrhoea, and other gastrointestinal and systemic disorders. Objective: This study amid to examines the antibiofilm activity of Laurus nobilis leaves extract on E. coli isolates and compares pre- and post-treatment gene expression of fimA and papC genes. Subjects and Methods: Ten isolates of E. coli were obtained from the Genetic Engineering and Biotechnology Institute, University of Baghdad, which was previously collected from Baghdad city hospitals and diagnosed by chemical tests, the diagnosis was confirmed using VITEK-2 System. The preparation of the aqueous and methanolic Laurus nobilis leaves extracts was done by using the maceration method and Soxhlet apparatus respectively. HPLC were conducted to determine the active compounds in the extracts. Moreover, molecular detection of fimA and papC genes and analysis of the gene expression by comparing the isolates treated with sub MIC of methanolic L. nobilis leaves extract with the untreated isolates. Results: Methanolic and aqueous extracts contained alkaloids, tannins, phenols, saponins, flavonoids, and glycosides. Seven polyphenolic compounds, four flavonoids derivatives (Apigenin, Luteolin, Rutin, and kaempferol) and three phenolic acids (Caffeic acid, Gallic acid, and Syringic acid), were identified by matching retention time with the standards. Laurus nobilis methanolic leaf extract inhibited 90% and 100% of E. coli biofilm development at 32 and 64 mg/ml. Conclusion: The result of the gene expression revealed that there is a decrease in the expression of the fimA and papC genes. The present study concluded that the Laurus nobilis leaves extract have rich phytochemical contents, so the methanolic extract had an excellent reduction effect on biofilm formation and showed remarkable down-regulation on the papC and fimA genes, which are responsible for the biofilm formation in E. coli.
A total of 165 clinical sample included Urine, Swab wounds and Burns were collected from Baghdad Governorate. Results showed that rate all isolates of E. coli was 50(30.3%) and rate of urine infection was 46(92%) and rate of swab wounds infection 4(8%). Where was diagnostic based on streaked on MacConkey agar, then single colony was transferred to Eosin Methylene Blue (EMB). Identification some of the biochemical test included: Catalase test, Oxidase test, Indole test, Methyl red, Vogues - Proskauer test and Citrate Utilization test. Then confirmed by the Vitek - 2 Compact System. The ability of E.coli isolate to biofilm formation to be studied it is considered one of the most important factors of virulence and has role in causing injury an

It has been demonstrated that Klebsiella pneumoniae produces a beta-lactamase that hydrolyses the β-lactam ring in antibiotics, making it one of the few bacteria which are currently exhibiting a high value of resistance because of changing in the organism's core genome. Methods: For the current study, 50 samples were gathered from different water sources, and based on morphological and biochemical testing, 10 isolates were determined to be K. pneumoniae. Accord

The Lactobacillus suspensions (L1 and L2) were individually and orally administered to experimental rats at a daily two consecutives of 100 μl (108 CFU/ ml/rat) for up to two weeks. Thereafter, on the 8th day of experiment rats were orally challenged with one dose infection of EHEC (105 CFU/ml/rat). Animals mortality and illness symptoms have been monitored. There was no fatal EHEC infection in rats that had been pre‑colonized with the Lactobacillus strains, while most of EHEC infected rats were died (90%). The

TheNative Americans and North New Mexico used a tea from the leaves and roots to treat asthma, headache, wound healing. As well as it was being consumed as daily dietary. All part of Y. gloriosa L. is rich in saponinsteroidal glycosides. Saponin extracts are well-known to be highly toxic. Hence, present study was carriedout to investigate the toxicity of saponin and estimate the LD50 value which helps in determining the safedose range for the drug that be used, as well as to determine hematological aspects and examine histologicaleffect. Different concentrations of saponin extract were injected into male mice (10,000, 8000, 6000, 400

Show MoreThis research was conducted to measure the safety of heat stable enterotoxin a (STa) produced by enterotoxigenic Escherichia coli, through studying its toxic effect on mice since it showed a promising effect in reducing the proliferation of colorectal cancer cells. The cytogenetic effect was determined after giving five different doses (100, 200, 400, 800 and 1600)μg/Kg in comparison with negative (phosphate buffer saline / PBS) and positive (mitomycin C/ MMC, at doses of 2 and 5μg/Kg) controls on mouse bone marrow cells by employing the following parameters: mitotic index, chromosomal aberrations and micronucleus, also, the serum level of liver functional enzymes (GOT, GPT, ALP) was recorded. In addition, lethal dose 50 (LD 50) with cert

Chemical phytochemical tests were performed for aqueous and alcoholic extracts of lemongrass. Antimicrobials activity was tested using agar disc diffusion method against Escherichia coli and Staphylococcus aureus. The results of the study showed that the aqueous extract of dried lemon leaves was highly effective (P≤0.05) against S. aureus, as the inhibition diameter was 22 mm for 50 dilution, while the inhibition diameter decreased to 15 mm for concentration 100. As for the alcoholic extract only, the diameter of inhibition decreased significantly (P≤0.0
... Show Moreraqi species of (Euphorbia lathyris F. Euphorbiaceae) have been used in the present study to evaluate genotoxicity of petroleum ether fraction in two doses (100mg/kg and 200mg/kg) on bone marrow cells in mice for seven successive days, comparing their effects with a positive control (methotrexate at a dose 20 mg/kg). Both doses of petroleum ether fraction of Euphorbia Lathyris significantly decrease mitotic index (P<0.05) compared to dimethylsulfoxide (negative control) group. For total chromosomal aberration, the petroleum ether fraction of Euphorbia lathyris (in both doses) produced significant increase (P<0.05) compared to dimethyl sulfoxide (negative control) group .at the same time, 20mg/kg methotrexate significantly increases (P<0.05)
