Extracellular Histones Inhibit Complement Activation through Interacting with Complement Component 4
Abstract
Complement activation leads to membrane attack complex formation, which can lyse not only pathogens but also host cells. Histones can be released from the lysed or damaged cells and serve as a major type of damage-associated molecular pattern, but their effects on the complement system are not clear. In this study, we pulled down two major proteins from human serum using histone-conjugated beads: one was C-reactive protein and the other was C4, as identified by mass spectrometry. In surface plasmon resonance analysis, histone H3 and H4 showed stronger binding to C4 than other histones, with KD around 1 nM. The interaction did not affect C4 cleavage to C4a and C4b. Because histones bin
... Show More

The relationship of eosinophil cationic protein / eosinophil count ratio and disease severity in allergic asthma patients

Finding an applicable clinical biomarker in body fluids can be useful in asthma management. This study was conducted to support the investigations of probable immunological changes in asthmatic patients that may be useful therapeutic indicators. 61 matching patients and 23 apparently healthy individuals were included. Significant elevation in the level of eosinophil cationic protein (ECP) in asthmatics was found as compared with healthy controls, so as among asthma severity categories. The receiver operating characteristic curve analysis determined 29 ng/ml of serum ECP as the typical cut-off value of significant denotation for differentiation between asthmatic patients and healthy individuals, significant positive linear cor

Investigation of the relationship between matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase with SARS CoV-2 infections

SARS-CoV-2 stands for severe acute respiratory syndrome coronavirus 2 which is the causative agent of spreading coronavirus disease 2019 that is known as COVID-19 pandemic, the disease leads to severe acute respiratory illness. Matrix metalloproteinases- 9 (MMP-9) plays several important physiological functions. This enzyme could also be implicated in the "cytokine storm" in some way, which may represent one of the possible scianrios during coronavirus infection, in addition to its role in the mechanism of lung fibrosis on molecular basis.. The tissue inhibitors of metalloproteinase (TIMPs) are well characterized for controlling the activity of MMPs in extracellular matrix remodeling. They also considered as signaling molecules anal
