Oak is considered a hard-to-root woody plant by conventional propagation methods, that’s why using tissue culture techniques is a very suitable alternative method. For oak embryo culture, WPM was used and found to be better than MS medium for embryo germination which gave 66.13%. The goal of the current work was to use Nigella sativa seeds' free and immobilised peroxidase to degrade textile dyes that pollute the environment and water. The enzyme was extracted for 15 minutes using a 1:20 (w:v) ratio, sodium acetate buffer at 0.2 M, and pH 5.0 after the optimal conditions for extracting the enzyme from Nigella seeds were determined. This yielded the highest specific activity of the enzyme, 1750 units/mg protein. The enzyme was purified in two stages: sucrose concentration and Sephadex G-150 gel filtration. This study was aimed to assess expression of IL-22 gene in patients with newly diagnosed autoantibodies against the thyroid antigens; anti-thyroglobulin (anti-TG), and anti-thyroid peroxidase (anti-TPO) in comparison with patients previously diagnosed and treated for autoimmune hypothyroidism as well as healthy control individuals. Twenty-eight patients with positive anti-TG and/or anti-TPO were enrolled in this study along with twelve age- and gender-matched healthy controls. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) was used to assess gene expression and fold change among the study groups.
