IL-18 Gene Polymorphisms Impacts on Its Serum Levels in Prostate Cancer Iraqi Patients

Prostate cancer is one of the most common types of cancer in men. A total of 110 Iraqi Arab individuals were included in this study; 60 individuals of them had prostate cancer with increased levels of TPSA (patients group); their age range 52-90 years. They were referred for diagnosis and treatment to the National Al-Amal Hospital for oncology in Baghdad during the period from July 2017 to October 2017. While the other 50 apparently healthy subjects were the control group, their age range similar to patients group. Sera and blood samples were collected from all patients and controls than used to assess for the level of IL-18 and DNA extraction, respectively. The polymorphisms were analyzed using polymerase chain reaction-single specific primer (PCR-SSP), at the position -137 G\C (rs187238) in the promoter of IL18 gene. The genetic polymorphism of the IL18 gene promoter -137G/C (rs187238) was determined and presented with three genotypes (GG, GC, and CC) in prostate cancer patients and controls. Testing for Hardy-Weinberg (H-W) equilibrium revealed that Prostate cancer patients showed insignificant variation in the distribution of IL-18 -137 genotypes (P> 0.05). While the control samples showed significant variation (p ≤0.05) between the observed and expected. Comparing patients with controls indicated that IL-18-137 alleles or genotypes showed no association with the risk of prostate cancer development in Iraqi Arab population or protection against them. Serum level of IL-18 was highly significant (P ≤ 0.001) increased in patients compared to control. The IL-18 serum levels differences in GG and GC genotypes was significant (p <0.05) between patients and control. While there were no significant differences between the three IL-18 -137 genotypes inpatient or in controls.

The Association Between IL-2 Gene (RS2069763 (Single Nucleotide Polymorphism and Type 2 Diabetes Mellitus in Iraqi Patients

This research attempts to find the association between single nucleotide polymorphism (SNP) of IL2+166 gene (rs2069763) and type 2 diabetes mellitus (T2DM) in a sample of Iraqi patients. A total of 44 patients and 55 apparently healthy volunteers were genotyped for the SNP using polymerase chain reaction test. Three genotypes (GG, GT, and TT) corresponding to two alleles (G and T) were found to have SNP. Both study groups’ genotypes had a good agreement for the analysis of Hardy-Weinberg Equilibrium. The results revealed increased frequencies between the observed and expected GG and TT genotypes and IL2+166 SNP T allele in T2DM  patients (40.9 vs. 40.0 %; OR = 1.04; 95% CI, 0.47 - 2.31)

Background: Cytokines are the messengers of the immune system.They are mostly secreted by macrophages and lymphocytes and their production is induced in response to injury or infections.
Objective: Biopsy speciemens from the middle body of the stomach were obtained from 18 patients with gastric carcinoma, 32 patients with atrophic gastritis, 50 patients with chronic gastritis and 20 healthy subjects and IL-8 and IL-1B levels were detrmined.
Methods: Sandwich-type enzyme-linked immunosorbent assay kit was used to determine the IL-8 and IL-1B levels.
Results: IL-8 and IL-1B levels were significantly higher in gastric carcinoma patients than patients with atrophic gastritis and both significantly higher than healthy subjects.

Relation- ships of neonatal septicemia with the mean serum levels of IL-8 and IL-1 in three large hospitals in Baghdad

Background: Neonatal septicemia (NNS) is the most serious complication in Neonatal Intensive Care Units (NICU) that demand urgent diagnosis and accurate treatment.
Methods: Serum was obtained from 31 neonates aged 1 hour-28 days that were diagnosed clinically and bacteriologically to have neonatal septicemia.
Results: Mean serum levels of both IL-8 and IL-1 recorded a significant increase in neonatal septicemia cases.
Conclusion: Usage of IL-8, IL-1 as diagnostic marker for NNS reduces unnecessary antibiotic therapy and therefore unnecessary costs, pain, and possible side effects of antibiotic
therapy and it may help to reduce development and spread of drug resistant bacteria.
