Determination of Vitamin B6 (pyridoxine hydrochloride) in Pharmaceutical Preparations Using High Performance Liquid Chromatography

Determination of vitamin B6 (pyridoxine hydrochloride) was described using high performance liquid chromatographic method. The analysis was achieved by cosmos IL 5C18-MS-II column (250 mm x 4.6 mm i. d., 5µm particle size) at room temperature. The mobile phase used was Acetonitrile, buffer solution (Citric acid, Na2HPO4 pH4) buffer solution in the ratio (70:30) (V: V). the flow rate was set to 1.25 mL.min-1 and the retention time 1.82 min with UV-detection at 282 nm. Beer's law was obeyed over the concentration range 10-1250 µg.mL-1. The method was accurate (relative error % less than 0.05%), precise (RSD better than ±1.05%), average recovery 100.05%, with a limit of detection and quantification of 2.2μg.ml−1, and 7.34μg.mL−1 respectively. The proposed method was successfully applied to determine the pyridoxine hydrochloride in pharmaceutical preparations in both forms of tablet and injection

Spectrophotometric Determination of Amiodarone Hydrochloride in Pharmaceutical Preparations

Amiodarone hydrochloride (AH) has been determined spectrophotometrically Using methyl orange (MO).  In our previous researches MO was used for determination of Mexiletine Hydrochloride [1]. The method based on complexation between MO and AH.  After shaking and diluting the complex solution with D.W, the pH was adjusted with NaOH and HCl to pH 3. The colored complex formed between AH and the reagent were transferred into separating funnels and extracted using 5.5ml CH2Cl2 and were shaken for (5 minutes).  The extracted organic layer was used for preparation of the calibration curves for spectrophotometric measurements of AH at 434nm.  The blanks were carried out in exactly the same way throughout the whole procedure.&n

The samples were first extracted using ethyl acetate and derivatized using ortho-phthaldialdehyde in the presence of 2-mercaptoethanol. C20 sphinganine was used as internal standard. Under the optimized conditions, separation was achieved using a mixture of methanol:water (93:7, v/v), column temperature at 30°C, flow rate of 1 mL min−1, and an injection volume of 10 μL. Chromatography was carried out on supelco L7 reversed- phase column (25cm × 4.6mm), 5 microns, using a mixture of methanol –acetonitrile-water: (80: 10 :10 v/v/v) as a mobile phase at a flow rate of 1.0 ml.min-1.
