Photodynamic Inactivation (Effect of Laser with and Without Photosensitizer) on Viability of Streptococcus Pyogenes

     This study focused on the bactericidal potency of toluidine blue” TBO”photosensitizer and red laser radiation of 635nmwith different doses against multi-drug resistant streptococcus pyogenes (S. pyogenes) isolated from infected burns wounds to see if it is susceptible to photodynamic inactivation .

Atotal of 45 isolates were collected from 38” patients” with infected burnwounds samples were collected from September to December 2019.Burns wounds swabs were employed using standard procedures of swab collection. Among these, eleven isolates were multidrug resistant”S.pyogenes”. More resistant isolates that has been proved to all antibiotics used. This multidrug resistant isolate used in all experiments. Bacterial suspension was diluted by using serial dilutions. The suspension of S.pyogenes in normal saline was treated with” red laser” radiation at a wavelength 635nm with and without “TBO”, and investigated the effect of changing laser doses (3.6 ,7 and 10.8 J/cm2) corresponding to laser exposure time (5,10 and 15 minutes  and different photosensitizer concentrations on viability of “S. pyogenes”isolated from infected burnswounds. The results of this study suggest that multi- drug resistance”S pyogenes” isolated from infected burn wounds inhibited by using photodynamic inactivation mediated by “TBO” and red laser at a wavelength 635 nm.  The effective technique for killing or inhibiting “S. pyogenes isolated from infected burns wounds is the combination of red laser at a wavelength 635nm and TBO. Thecombination of TBO and red laser light prohibit”S. pyogenes”, the optimum results of bacterial inhibition obtained at 50µg/ml, and laser dose 7 J/cmcorresponding to exposure time 10 minutes.

The Effect of Diode Laser on Viability and Antibiotic Sensitivity of Streptococcus mutans Isolated From Dental Caries

Streptococcus mutans is one of the major cariogenic microbial flora. In an attempt to determine the mutagenic effect of diode laser on the viability and antibiotic sensitivity of this bacteria; A total of 30 samples were collected from dental caries. The isolates were identified using by conventional identification methods and confirmed using VITEK2 system. The Inhibition of Streptococcus mutans by He- Ne Laser via TBO Photosensitizer

This work aims to investigate the inhibition of vitality of Streptococcus mutans, which is the causative agent of caries. A 632.8 nm He-Ne laser with the output power of 4.5mW was used in combination with toluidine blue O (TBO) at the concentration of 50μg/ml as a photosensitizer. Streptococcus mutans was isolated from 35 patients if carious teeth. Three isolates were chosen and exposed to different energy densities of He – Ne laser light 3.8, 11.7, 34.5 and 104.1 J/cm². After irradiation, substantial reduction was observed in the number of colony forming units (CFU)/ ml. The reduction in the number of CFU was increasing as the dose increased.

Photodynamic Inactivation of Candida Albicans Sensitized by Malachite Green

The aim of this study is to investigate the ability of malachite green (MG) combined with 650nm diode laser to kill Candida albicans and to spectrally study the MG photodegradation after photodynamic therapy (PDT) spectrally. Cultures of Candida albicans were exposed to 40mW, 650 nm diode laser in the absence of MG. In PDT group, the MG was added to the Candida suspension for 5 min then exposed to diode laser for (5, 10, 15, 20) min at power density of 0.59W/cm2. Purification and characterization of Streptococcus pyogenes superantigen (Spe-C)

From 144 specimens of tonsillitis which were collected from patient, (children of 3 -12 year olds) there were 70 isolates beta hemolytic and 28 isolates were identified as S. pyogenes. Sensitivity of S. pyogenes isolates to antibiotics was tested, all isolates were sensitive to amoxicillin and cephaloxia while higher resistant were to erythromycin. One isolate whiche was 100 A had a stable characteristics and produce pyrogenic toxin was chosen for study and it was purified and characterized from the cell free supernatant of S. pyrogenes strain.

The Effect of He:Ne Laser on Viability and Growth Rate of Leishmania Major

An isolate of Leishmania major was grown on the semisolid medium and incubated at 26ºC. The isolate was irradiated by He: Ne laser (632.8 nm, 10 mW) at exposure times (5, 10, 15, 20, 25, 30) minutes in their respective order. The unirradiated groups represent control group.
