Analysing Aflatoxin Production Conditions in Feed Samples Using a Preparative Thin Layer Chromatography (TLC) Method

    In this research, aflatoxins were produced, extracted, isolated, and purified in order to optimize the storage conditions of feed. Using a preparative thin layer chromatography (TLC) method, with commercially available plates of 1.5 to 2.0 mm, B1 aflatoxin was isolated from the feed samples of whole wheat, maize, and crushed rice, and the procedure was repeated four times. A purity value of 99 % for B1 aflatoxin was achieved and tested using spectrophotometric and chromatographic methods. As solvents, acetone:water (85:15l) was used for aflatoxin extraction from the feed sample, whereas methanol: water (50: 50) was used for trichothecenes extraction. The primary findings of this research indicate that B1 aflatoxin reached a maximum yield when coarsely crushed rice was inoculated with  Aspergillus parasiticus at a temperature of 30C°, moisture level of 35%,  and incubation duration of 7 days. The observations obtained from aflatoxins production process provide an insight regarding storage conditions that need to be avoided in order to reduce the incidents of feed contamination.

Production of Slime Layer by Staphylococcus epidermidisIsolated From Corneal Infection

A total of 37 Staphylococcus epidermidis isolates, isolated from corneal scraping of patients with bacterial keratitis and 20 isolates from healthy eyes (as control) (all isolates, isolated from, Ibn Al- Haietham eye hospital / Baghdad), were tested for slime production, 52.63% of all isolates were positive-slime production (23 isolates from patients and 7 isolates from controls). It was found that positive-slime producing S. epidermidis were exhibited a high resistance to antibiotics as compared to negative-slime producing isolates.

Direct Acetylation and Determination of Chlorophenols in Aqueous Samples by Gas Chromatography Coupled with an Electron-Capture Detector

A method was developed that offers a rapid, simple and accurate technique for the determination of chlorophenols at trace levels in aqueous samples with very limited volumes of organic solvents. These compounds were acetylated, then preliminarily extracted with n-hexane. The enriched chlorophenols were directly analyzed using gas chromatography with an electron-capture detector. The detection limits were in the range of 0.001–0.005 mg/L, except for 2-chlorophenol, which was always above 0.013 mg/L. Relative standard deviation for the spiked water samples ranged from 2.2 to 6.1%, while relative recoveries were in the range of 67.1 to 101.3%.

Analysing of Hueckel Edge Detector Performance Using Satellite Image

In this research, an analysis for the standard Hueckel edge detection algorithm behaviour by using three dimensional representations for the edge goodness criterion is presents after applying it on a real high texture satellite image, where the edge goodness criterion is analysis statistically. The Hueckel edge detection algorithm showed a forward exponential relationship between the execution time with the used disk radius. Hueckel restrictions that mentioned in his papers are adopted in this research. A discussion for the resultant edge shape and malformation is presented, since this is the first practical study of applying Hueckel edge detection algorithm on a real high texture image containing ramp edges (satellite image).

Optimum conditions of keratinase production from Bacillus licheniformis

Forty isolates of Bacillus spp. were isolated from fifty samples including different source of soil to detect the ability to produce keratinase enzyme in liquid state fermentation, Bacillus (Bs13)was the highest keratinase producer , it was identified as a strain of Bacillus licheniformis. The optimum conditions for keratinase productions were in a media contains keratin 4% (hooves) as a carbon and nitrogen and energy sources, peptone 1% as a secondary nitrogen source with pH 8 , inculums size 1%, and incubated at 37Co for 24 hrs.

Optimum conditions for fibrinolytic enzyme (Nattokinase) production by Bacillus sp. B24 using solid state fermentation

The present study was aimed to screen the ability of local isolates of Bacillus spp. (56 isolates) for nattokinase production using solid state fermentation, then optimize the nutritional conditions for enzyme production.
