Materials and Methods Bacterial strains P. aeruginosa was obtained from postgraduate students Laboratories of Biology Department/College of Science/University of Baghdad. That previously isolated from patient suffering from Cystic Fibrosis. API 20 NE system was employed for the identification of P. aeruginosa. A total of 122 urine specimens were collected in the period between of mid of July until to the mid of September of 2010 from AL-Kadhmiya Teaching Hospital in Baghdad City. Specimens were collected from out-patients in sterile screw cupped containers. Regarding inpatients, catheter was withdrawn and cut
Bacteria form complex and highly elaborate surface adherent communities known as biofilms.Biofilm have been shown to be associated with several human diseases ,and to colonize a wide variety of medical devices . The current study focuses on contribution of extracted genomic DNA in biofilm formation by P. aeruginosa and K. pneumoniae isolates .The percentages of Pseudomonas aeruginosa recovery from drinking water in this study were 10%(20 positive P. aeruginosa samples ) and K. pneumonia., 7%(14 positive K. pneumonia samples). The results showed that all P.aeruginosa and K. pneumoniae isolates (100%) were slime producer but in different degrees by forming of black
... Show MoreThe study was aimed at inhibiting the protease produced by Pseudomonas aeruginosa using an 80% alcoholic extract of Conocarpus lancifolius leaves. A total of 146 isolates of P. aeruginosa that were isolated and identified by microscopic and biochemical tests were 51 isolates submitted to primary and secondary screening techniques in order to choose the qualified P. aeruginosa isolate for protease synthesis. Among these isolates, forty-seven isolates showed hydrolysis zones on skim milk media (primary screening); six isolates were chosen for secondary screening. The result revealed that P. aeruginosa P51 had the highest ability to produce the enzyme, with a specific activity of 15.9 U/
... Show MorePseudomonas aeruginosa, a ubiquitous environmental organism, is a difficult-to-treat opportunistic pathogen due to its broad-spectrum antibiotic resistance and its ability to form biofilms. In this study, we investigate the link between resistance to a clinically important antibiotic, imipenem, and biofilm formation. First, we observed that the laboratory strain P. aeruginosa PAO1 carrying a mutation in the oprD gene, which confers resistance to imipenem, showed a modest reduction in biofilm formation. We also observed an inverse relationship between imipenem resistance and biofilm formation for imipenem-resistant strains selected in vitro, as well as for clinical isolates. We identified two clinical isolates of P. aeruginosa from the sputum
... Show MoreThis study aimed to explore whether green synthesized copper nanoparticles (CuNPs) can function as an anti-biofilm agent produced by P. mirabilis. The nanoparticles were synthesized from cells free extract of P. mirabilis. Characterization of biosynthesized copper nanoparticles was carried out to determine the chemical and physical properties of the product using atomic-force microscopy (AFM), transmission electron microscopy (TEM), field emission scanning electron microscopy (FESEM), X-ray diffraction (XRD) and UV-visible spectroscopy. The hexagonal structure was confirmed by XRD, size range was marked 13-19nm by TEM. FESEM was used to confirm the surface morphology. AFM analysis was used to reveal the roughne

Microalgae present much usefulness for antimicrobial research because of its enormous biodiversity and rapid growth rate. From this study results it is reaveled that Chlamydomonas reinhardtii were isolated from a pond of water in the province of Diwaniyah. The culture supernatants were obtained when extracted with methanol solvent. Antimicrobial activity of extracts was tested for pathogens, and the best inhibition zone obtained was against Candida albicans (32mm), S.aureus (15mm), and to E.coli (9mm). While it showed no effect against both S.epidermidis and Klebsiella spp. Biofilm was formed by all tested isolates with differences in its strength formation. The C. reinhardtii

producers and biofilm formation. The activities of two selected K. pneumoniae
isolates for their biofilm formation and susceptibility to antibiotics after treatment
with several plants extracts were investigated. The results of water and 60% ethanol
extracts for Matricaria chamomile flowers, Alhagi maurorum leafs, Syzygium
aromaticum buds (clove) and Arctium minus leafs were showed reduction of biofilm
Eight isolates of P. aeruginosa were obtained out of 90 water samples. The isolated colonies were identified based on their morphology and biochemical characteristics, were confirmed as P. aeruginosa by the API 20E test system.
The percentages of P. aeruginosa recovery in this study were 8.8.% All isolates were able to produce greenish blue pigment (pyocyanin). Pyocyanin at all concentrations was significantly increased the percentage of fragmented DNA of peripheral blood lymphocyte cells compared to control , results showed that DNA fragmentation percentage was higher in concentration 50 μg/ml (70%,74.3%) at 24 hr,48hr respectively. In summary, results of recent study demonstrate that the pyocyanin, induces apoptosis of human periph
In this paper, quantified study of the biofilm formed by Klebsiella pneumoniae isolated from urine specimen of patient suffering from acute urinary tract infection (UTI) on catheter, stainless-steel and glass coupon surfaces, as well as determine the relationship between time contact and biofilm progression using crystal-violet binding assay based on the values of optical density at 620nm of the crystal violet stain which bonded total biofilm biomass by resolubizing with 99.9% ethanol at the specific interval times. The result showed biofilm formed on three tested surfaces but in different degrees. According to obtained data, the catheter coupons presents a higher capability to attract bacteria cell and biofilm formation followed by glas

For this purpose, 1% and 2% concentration of each of glucose and sodium chloride were used to test the biofilm formation potential of Staphylococcus aureus and Pseudomonas aeruginosa, which were the most common abundant bacteria that cause infection by biofilm. Each of the concentrations was kept in contact with the pathogenic bacteria for 24 hours. After the period of incubation, the concentration of 1% of glucose enhanced moderate biofilm formation capacity for (66% and 80%) on both bacteria respectively. The concentration of 2% glucose, on the other hand, led to a weak biofilm fo
